Single-cell genomics has transformed our understanding of cellular heterogeneity, but the regulatory mechanisms that govern cell identity and function are often shaped by tissue architecture and local microenvironmental interactions that are lost during tissue dissociation. Recent advances in spatial epigenomic and multi-omic technologies leverage genome-wide, unbiased sequencing approaches to profile chromatin accessibility, histone modifications, DNA methylation and gene expression directly within intact tissues. By capturing regulatory information beyond RNA expression alone, these technologies enable researchers to investigate gene regulation, cellular states and tissue organisation in their native spatial context.

In this seminar, Dr Chen will introduce Spatial ATAC-seq, Spatial CUT&Tag and emerging spatial multi-omic workflows, highlighting applications in cancer, neuroscience, development and ageing research. Practical considerations for incorporating spatial epigenomic and multi-omic profiling into biological research will also be discussed.
