Keeping mitochondria healthy depends on tight quality control, and ubiquitination has emerged as a central mechanism governing mitochondrial dynamics, protein import into mitochondria, mitophagy, and stress responses. In this seminar I will discuss how ubiquitin writers (E3 ligases), erasers (deubiquitinases) and a special ubiquitin modifier (PINK1) mark mitochondria for degradation or repair. This system of ubiquitin control offers an attractive therapeutic strategy for modulating mitochondrial function. In line with this, I will discuss some examples in which the ubiquitin system is being harnessed to treat neurodegenerative disease.
