Ruxolitinib, which is currently in clinical use for MPN patients, may act as a pathogenic signaling node released upon drug withdrawal, especially in JAK2V617F patients. Ultra-precision medicine approach tailored to either Type 1 OR Type 2 mutation in CALR driven MPN patients will be required for maximal efficacy, with far-reaching implications for patient management.

 

Denis Tvorogov is an international expert on cytokine receptor signalling having trained with two of the leading receptor tyrosine kinase researchers in the world, Graham Carpenter in Nashville, and Kari Alitalo in Helsinki. He is highly skilled in molecular biology and cell membrane biochemistry with expertise in the mechanisms of cytokine receptor dimerization and tyrosine kinase signalling. He has a strong focus on output quality as exemplified by publications in Cancer Cell, Blood,  Sci. Advances, Nature and Cancer Discoveries . His previous expertise in antibody therapy and intimate knowledge of cytokine receptor oligomerisation-initiated signalling has recently led to exceptional and paradigm shifting discoveries (Tvorogov et al. Sci.Advances, Cancer Cell) that took advantage of structural biology approaches (Kan et al., Cancer Discoveries). Denis Tvorogov has uncovered the molecular mechanisms underpinning ruxolitinib withdrawal syndrome and ruxolitinib resistance published in Science Advances in 2018 and proposed first specific antibody base therapy for CALR-driven MPNs (Tvorogov et al Embo Reports 2022, Thompson-Peach et al, Blood 2026).
