Interrogating dynamic Bcl-2 protein interactions at mitochondria

My broad area of interest is around understanding how cells detect and respond changes in their extracellular matrix (ECM) microenvironment. We use a range of approaches, from studying tissue composition in clinical trials to cell culture models. One aspect of particular interest to us is how signals from the ECM alter apoptotic priming at mitochondria and the consequences of this for epithelial cell function. We have a longstanding interest in how adhesion receptor mediated signalling alters Bcl-2 protein dynamics the mitochondrial membrane and ow this regulates priming. To understand the mechanisms that might regulate the dynamic exchange of Bcl-2 proteins between subcellular compartments we have employed proximity-dependent biotin identification (BioID). Comparing BioID using a range of Bcl-2 family bait proteins, we highlight a complex range of interacting partners on membranes that can vary dynamically with priming.
