Technology

The Screening Laboratory supports three fully automated liquid handling systems and various types of plate readers, enabling high capacity assays to be formatted using a wide range of technologies. The range of assay technologies supported for formatting molecular screens includes

  • AlphaScreen™

  • Time resolved fluorescence

  • Fluorescence polarization

  • Steady-state fluorescence

  • Photometry

  • Chemiluminescence

  • ELISA

  • Scintillation proximity

  • Filtration-based separations

  • Surface plasmon resonance (SPR) using the Biacore 4000

For cell-based screens, supported assays include:

  • Reporter genes (luciferase, green/yellow fluorescent proteins, alkaline phosphatase, beta-galactosidase)

  • BRET™ (protein-protein interactions)

  • Cell-surface antigen expression

  • Cytokine expression

  • Cell proliferation and cytotoxicity

Libraries

  • The WECC library is composed of approximately 114,000 “lead like” small chemical entities. This library is our screening starting point to discover new hit compounds.

  • The epigenetic library is composed of 95 compounds described to interact with epigenetic targets

  • The kinase library is composed of 276 compounds targeting kinases

  • Targeted agents library

  • The known drug library is composed of 3,700 developed drugs that are on the market or failed during clinical trial

  • The malaria box library is composed of 400 compounds that have shown activity against the asexual blood stage of P. falciparum.
