Research Overview

There is good evidence that interfering with the Bcl-2-regulated pathway to cell death is a promising therapeutic approach. The race is now on to find small molecules, dubbed as “BH3-mimetics”, capable of either killing cancer cells directly or in combination with common chemotherapeutic drugs by acting on the Bcl-2-regulated pathway. Results emanating from our Division have shed light on the selective binding properties of the BH3-only proteins to pro-survival members of the Bcl-2 family, and shown the need for a better understanding of the properties and mechanisms of regulation of the BH3-only proteins.

Our present work is aimed at evaluating the importance in vivo of mechanisms of regulation that have so far been discovered and characterised only in vitro. Bim is the primary focus of our studies, because it plays a prominent role in the haematopoietic system, and is readily amenable to gene targeting. We have generated several mouse strains in which the Bim gene contains subtle mutations that address different mechanisms of regulation.
