WEFCS - For Flow Cytometry Data Analysis On VAX Systems

The techniques for flow cytometry data analysis have been brought together into a single program to run on Vax computer systems. Histogram, two-parameter "contour", or list mode data files of up to 8 flow cytometric parameters can be analysed and appropriate conversions between these three data formats are possible. For list mode data, both histogram displays and two-parameter correlated displays are constructed simultaneously on a single reading of the file. Graphics input cursors may then be used in either of the display formats for setting analysis gates or for extraction of numeric data (e.g. quadrant analysis, sub-population proportions, percentage positives, etc.). Other features include probability density calculation, simple DNA curve fitting, negative control background subtraction and a more flexible formatting of figures to be laser printed for publication.

The Fortran program was developed to run on the WEHI microvax cluster but has very little machine specificity. The cluster includes a large data storage volume which receives data from FACScan/Consort30, FACStar+/ConsortVax, and FACSII/pdp11/73 flow cytometers. The program was therefore designed to read the different data and file header formats written by these three acquisition systems as well as reading the flow cytometry standard format (FCS 1.0). WEFCS may be run by several operators simultaneously using any of a number of graphics display terminals, personal computers with terminal emulation, or via a Tek window on a Vaxstation console. Similarly, monochrome graphics paper hard copy can be requested on any of a number of laser printers.
