Lymph Node; Forward Scatter linking to 2D display [1.02MB]

FS-H (forward light scatter; a measure of cell size) links to FL1-H (CD3) vs. FL2-H (B220). Watch for: 1. Mostly single-positives at low FS. 2. Double-positives dominate at FS greater than about 140 units. 3. The small population of double-negatives are maximal at FS about 100-140.

Lymph Node; 2D display linking to Forward Scatter [1.37MB]

FL1-H (CD3) vs. FL2-H (B220) links to FS-H (forward light scatter). Watch for: 1. B220 single-positive are smaller in FS than CD3 single-positive. 2. The double-positives are huge in FS. 3. The small population of double-negatives have intermediate FS.

FS-H (forward light scatter) vs. SS-H (Side scatter; a measure of nuclear asphericity) links to FL1-H (Mac-1) vs. FL2-H (CD45RA) and to FL3-H (Class II) vs. FL4-H (CD11c) Watch for: Small population that appears to be moderate FL1+/FL2+ and FL3+/FL4+, possibly autofluorescent macrophages, materializes at very specific FS/SS position.

Dots are colour-coded for cluster membership. Watch for: The small purple population in the 3D display looking mildly FL5-positive is a small remnant of dead cells.
